Age- and quality-dependent DNA methylation correlate with melanin-based coloration in a wild bird ArchiMer
Soulsbury, Carl D.; Lipponen, Anssi; Wood, Kristie; Mein, Charles A.; Hoffman, Joseph I.; Lebigre, Christophe.
Secondary sexual trait expression can be influenced by fixed individual factors (such as genetic quality) as well as by dynamic factors (such as age and environmentally induced gene expression) that may be associated with variation in condition or quality. In particular, melanin-based traits are known to relate to condition and there is a well-characterized genetic pathway underpinning their expression. However, the mechanisms linking variable trait expression to genetic quality remain unclear. One plausible mechanism is that genetic quality could influence trait expression via differential methylation and differential gene expression. We therefore conducted a pilot study examining DNA methylation at a candidate gene (agouti-related neuropeptide: AgRP) in...

Alelle number and heterozigosity for microsatellite loci in different stingless bee species (Hymenoptera: Apidae, Meliponini) Neotropical Entomology
Francisco,Flávio de O.; Brito,Rute M.; Arias,Maria C..
In the present study we compare genetic characteristics (allele diversity and observed heterozygosity) of microsatellite loci, from three stingless bee species (Plebeia remota Holmberg, Partamona mulata Moure In Camargo and Partamona helleri Friese), amplified by using heterospecific primers originally designed for Melipona bicolor Lepeletier and Scaptotrigona postica Latreille. We analyzed 360 individuals of P. remota from 72 nests, 58 individuals of P. mulata from 58 nests, and 47 individuals of P. helleri from 47 nests. The three species studied showed low level of polymorphism for the loci amplified with primers derived from M. bicolor. However, for the loci amplified with primers derived from S. postica, only P. remota presented low level of...

Allozyme variation in a natural population of Stryphnodendron adstringens in the Rio Preto State Park, southeastern Brazil Rev. Bras. Bot.
Glasenapp,Jacqueline Siqueira; Barbosa,Priscila Barros; Casali,Vicente Wagner Dias; Martins,Ernane Ronie; Cruz,Cosme Damião.
Leaves and fruits from 63 Stryphnodendron adstringens trees were sampled in the Rio Preto State Park to analyze allozyme segregation, tissue specific expression of allozyme loci, and their genetic parameters. The enzyme systems ADH, EST, ACP, PGM, PGI, GDH, G6PDH, GOT, IDH, LAP, MDH, PER and SKDH were assessed by means of starch-gel electrophoresis. The polymorphic systems PGI, IDH, MDH and GOT demonstrated a dimeric quaternary structure, while EST and PER were monomeric. The total expected genetic diversity (H E) for leaves and seeds were 0.325 and 0.244 respectively. The effective number of alleles per locus (A E) was 1.58 in leaves and 1.42 in seeds. The values of H E and A E observed in S. adstringens were comparatively higher than the average values...

Characterization of 13 microsatellite loci developed from Meconopsis horridula Genet. Mol. Biol.
Zhao,Yu; Zhang,Shi-Bao; Yang,Jing; Zhang,Lu.
Meconopsis horridula is one of the eight most famous flowers in Chinese province of Yunnan. In this study, a modified biotin-streptavidin capture method was used to detect 13 microsatellite markers in the genome of M. horridula. The polymorphism of each locus was assessed in 24 samples collected from four populations. The number of alleles per locus ranged from 2 to 7 (mean: 3.2). The observed and expected heterozygosities ranged from 0.0833 to 0.9167 and 0.0816 to 0.8050, respectively. Additionally, nine of the 13 microsatellite markers were successfully amplified in three other congeneric species. These polymorphic SSR markers could be useful for studying the population genetics of M. horridula and for assessing genetic variation in this and congenerc...

Vela Avitúa,Sergio; Montaldo,Hugo H; Márquez Valdelamar,Laura; Campos Montes,Gabriel R; Castillo Juárez,Héctor.
Background: The objective of this study was to estimate the decline of genetic variability and the changes in effective population size in three shrimp populations. One was a wild population collected at several points in the Mexican Pacific Ocean. The other two populations were different generations (7 and 9) from a captive population selected for growth and survival. Microsatellite markers and pedigree were both used to assess genetic variability and effective population size. Results: Using 26 loci, both captive populations showed a decline in the expected heterozygosity (20%) and allelic diversity indices (48 to 91%) compared to the wild population (P < 0.05). The studied captive populations did not differ significantly from each other regarding...

Genetic diversity and environmental stress in Italian populations of the cyprinodont fish Aphanius fasciatus ArchiMer
Cimmaruta, R; Scialanca, F; Luccioli, F; Nascetti, G.
Allozyme electrophoresis has been used to analyse the genetic structure of ten populations of the killifish, Aphanius fasciatus, sampled from Italian coastal brackish lagoons or salterns. The goal of the study was to compare the genetic structure of samples obtained from habitats characterised by environmental stress of different kinds, with the aim of gaining a better understanding of the relationships between the genetic variation of natural populations and environmental disturbance, either natural or man-made. Therefore, the sampling scheme has been planned to provide samples from both brackish habitats, characterised by natural stress with seasonal marked fluctuations of chemical and physical parameters, and from coastal lagoons suffering from a high...

Genetic diversity of 16S rRNA and mcrA genes from methanogenic archaeas Biological Sciences
Xavier, Keyla Vitoria Marques; Souza, Eden Silva e; Santos, Erick de Aquino; Diniz, Michely Correia.
Methanogenic archaeas are found in aquatic and terrestrial environments and are fundamental in the conversion of organic matter into methane, a gas that has a potential use as renewable source of energy, which is also considered as one of the main agents of the greenhouse effect. The vast majority of microbial genomes can be identified by a conservative molecular marker, the 16S ribosomal gene. However, the mcrA gene have been using in studies of methanogenic archaea diversity as an alternative marker, highly conserved and present only in methanogens. This gene allows the expression of the enzyme Methyl-coenzyme M reductase, the main agent in converting by-products of anaerobic digestion into methane. In this context, we aimed to study the genetic...

Genetic diversity of Brazilian triticales evaluated with genomic wheat microsatellites PAB
Costa,Cibele Tesser da; Albuquerque,Ana Christina Sagebin; Nascimento Junior,Alfredo do; Marcelino,Francismar Correa; Pereira,Jorge Fernando.
The objective of this work was to determine the genetic variability available for triticale (X Triticosecale Wittmack) crop improvement in Brazil. Forty-two wheat genomic microsatellites were used to estimate the molecular diversity of 54 genotypes, which constitute the base of one of the major triticale breeding programs in the country. Average heterozygosity was 0.06 and average and effective number of alleles per locus were 2.13 and 1.61, respectively, with average allelic frequency of 0.34. The set of genomic wheat microsatellites used clustered the genotypes into seven groups, even when the germplasm was originated primarily from only two triticale breeding programs, a fact reflected on the average polymorphic information content value estimated for...

Genetic structure of bisexual and parthenogenetic populations of Artemia from Italian brackish-hypersaline waters ArchiMer
Nascetti, G; Bondanelli, P; Aldinucci, A; Cimmaruta, R.
Eight populations of Artemia displaying different reproduction modalities (amphigonic vs. parthenogenetic) and ploidies (diploid vs. polyploid) have been collected from Italian salt-works or brackish coastal lagoons, and their genetic structure has been investigated using allozyme electrophoresis. The results obtained on genetic differentiation showed that the Tyrrhenian bisexual samples (A. salina) are genetically homogeneous even between populations located in relatively faraway sites (Sardinia-Latium), having a moderate gene flow assisted by migratory waterfowl. The parthenogenetic samples (A. parthenogenetica) are genetically well differentiated both from the bisexual ones and among them, even when characterised by the same ploidy. Each sample presents...

Genetic variability and evidence of founder effect in Hemiodus orthonops (Characiformes: Hemiodontidae) from the upper Paraná River basin, Brazil Biological Sciences
Marques, Liégie Alher; Renesto, Erasmo.
Hemiodus orthonops is a small fish of the Hemiodontidae family, order Characiformes, with a maximum of 25 cm standard length. Until recently, H. orthonops was an endemic species from the Paraná-Paraguay basin and it was absent from the upper Paraná River basin. Since 2008, it has started to be collected in the upper Paraná River, representing up to 10% of catches. Two population samples of H. orthonops from two localities of the upper Parana River basin (Porto Camargo and Porto Figueira) were analyzed using the allozymes electrophoresis technique. Twenty-one enzymatic loci were detected. The population sample from Porto Camargo displayed a genetic variability (He = 0.1061) higher than that from Porto Figueira (He = 0.0580) and homozygote excess in both of...

Paiva,Suzana de; Renesto,Erasmo; Zawadzki,Cláudio Henrique.
Hypostomus strigaticeps and two morphotypes of Hypostomus were collected from Ribeirão Maringá, a small tributary of the Rio Pirapó, an effluent of the upper Rio Paraná. The three populations were analyzed by allozyme electrophoresis that allowed the scoring of 25 loci from 14 enzyme systems. Heterozygosity values (He) were 0.028 in H. strigaticeps, 0.027 in Hypostomus sp. 1 and zero in Hypostomus sp. 2. Several diagnostic loci and fixed differences were observed for each population at loci Acp-A, Gcdh-A and Mdhp-A. Thus, all populations were genetically distinct, although there were many common alleles. The unbiased genetic identities of Nei (I) were estimated as 0.780 for Hypostomus sp. 1 and H. strigaticeps, 0.357 for H. strigaticeps and Hypostomus sp....

Genetic variability of native populations of Wasmannia Forel (Hymenoptera: Formicidae) and their biogeographical implications Neotropical Entomology
Souza,Ana L B; Tavares,Mara G; Serrão,José E; Delabie,Jacques H C.
Wasmannia auropunctata (Roger) and Wasmannia rochai Forel are economically important ants in the Southeast and Southwest regions of Bahia State, Brazil. Randomly Amplified Polymorphic DNA (RAPD) markers were used to distinguish and analyze the genetic variability of populations of both species from Ilhéus, Jequié, BA and French Guyana. The genetic distances among W. auropunctata and W.rochai populations (55.8-71.4%) suggest genetic differentiation among them. Wasmannia auropunctata populations from sugarcane and banana plantations in Jequié were the most distant genetically (30.1-46.3%) and may represent populations restricted to isolated fragments. The high genetic distances among W. auropunctata populations from CEPLAC experimental areas, in Ilhéus...

Genotypic characterization of microsatellite markers in broiler and layer selected chicken lines and their reciprocal F1s Scientia Agricola
Rosário,Millor Fernandes do; Ledur,Mônica Corrêa; Moura,Ana Silvia Alves Meira Tavares; Coutinho,Luiz Lehmann; Garcia,Antonio Augusto Franco.
Chicken experimental populations have been developed worldwide for QTL mapping, but their genotypic characterizations are not usually discussed. The objective of this study was to characterize genotypically two F1 reciprocal generations and their parental lines based on the estimation of genotypic parameters. These F1 generations originated two Brazilian reference populations to map QTL. The evaluated parameters were polymorphic information content (PIC), observed and expected heterozygosities and number of alleles at microsatellite loci on chromosomes 1, 3 and 4. All parental and F1 chickens from both populations were used totalling of 83 chickens: 14 from a broiler (TT) and 14 from a layer line (CC) and 55 from their reciprocal F1 generations. The...

Heterozygosity and hammuli number asymmetry in adult workers of Apis mellifera (Hymenoptera, Apidae). Relationship between individual heterozygosity at particular loci (Mdh-1, Hk-1 ou Pgm-1) or average heterozygosity of the colony and asymmetry of the hammuli number of right and left wings was verified on adult workers of five Africanized and five European colonies of Apis mellifera. Our results demonstrated the importance of statistical tests to verify the type of asymmetry of the character studied and they did not validate the assumption that uni or multiloci heterozygosity has a positive effect on developmental stability.

Microsatellite-associated heterosis in hatchery-propagated stocks of the shrimp Penaeus stylirostris ArchiMer
Bierne, Nicolas; Bezuart, Ivan; Vonau, Vincent; Bonhomme, François; Bedier, Edouard.
Correlation between DNA microsatellite heterozygosity and growth rate was investigated in two hatchery-propagated stocks of the shrimp Penaeus stylirostris, which had been genetically isolated from wild founders for 17 generations, and from each other for five more generations. presumed demographic history of these populations suggested that they were maintained at small effective population sizes. and this was confirmed by significant changes in allelic frequencies between the two stocks. Despite a small sample size (n = 60) and a small number of loci screened, a significant positive correlation was detected between microsatellite tri-locus heterozygosity and growth rate in one of the two stocks (r = 0.3, P = 0.02). In the other stock (n = 48), the...
